PolyCYPs® Enzymes
PolyCYPs® enzymes are mined from Hypha’s talented microbial biotransformation platform and capable of oxidising a wide range of substrate compounds, especially arylic/benzylic -CH, -CH2, -CH3, -tert-butyl, and iso-propyl moieties, as well as aromatic systems for phenols and epoxides. De-alkylation of N- and O-alkyl moieties is also observed.
These enzymes have been successfully used by pharma and agrochemical clients to create scalable amounts of mammalian CYP-derived metabolites, as well as oxidised late-stage derivatives of drugs. Clients have also made use of the option to outsource screening and scale-up PolyCYP reactions to Hypha.
PolyCYPs+ Metabolite Screening Kits
PolyCYPs® enzymes are formatted into a screening kit together with aldehyde oxidase and flavo-containing monooxygenase 3, which allows the convenient and cost effective in-house testing of compounds for scalable synthesis of many oxidised drug metabolites. PolyCYPs+ kits contain 18 PolyCYPs enzymes with human AOX1 and FMO3.
Reactions are scalable either by resupply of the enzymes for production in-house, or for larger scale, production and purification is available at Hypha.
Enzymes in Hypha’s oxidation kit have been rigorously tested for biocatalytic ability against a diverse panel of substrates. So far 87% of a panel of 50 CYP substrates tested have been biotransformed by PolyCYPs enzymes to at least one metabolite.

Case study illustrating rapid scale-up of a hydroxylated metabolite using PolyCYPs. A US pharma client wished to quickly access milligram quantities of a hydroxylated drug metabolite. Using PolyCYPs, Hypha were able to synthesize and purify the metabolite for the client within 3 weeks, together with a Certificate of Analysis. Read about this project here.
